[摘要]
目的:研究神經(jīng)生長(zhǎng)因子(Nerve growth factor, NGF)對(duì)人血管內(nèi)皮細(xì)胞(Human vascular endothelial cell, HEVC)增殖作用的影響。
方法:將NGF作用于體外培養(yǎng)的HEVC304,利用酶聯(lián)免疫反應(yīng)檢測(cè)其增殖率,利用免疫組織化學(xué)的方法檢測(cè)對(duì)增殖細(xì)胞核抗原(Proliferating cell nuclear antigen, PCNA)的表達(dá)和標(biāo)記指數(shù)。
結(jié)果:NGF作用組較對(duì)照組其細(xì)胞增殖率明顯增加(P<0.001), NGF中和抗體組其細(xì)胞增殖率下降明顯(P<0.05)。NGF作用組較對(duì)照組能明顯增加HEVC對(duì)PCNA的表達(dá)和PCNA標(biāo)記指數(shù)(P<0.05), NGF中和抗體組其PCNA表達(dá)和PCNA標(biāo)記指數(shù)明顯減少(P<0.05)。
結(jié)論:NGF能明顯促進(jìn)HEVC的增殖。
Study of effect of nerve growth factor on the human vascular endothelial cell proliferation Abstract: Objective To investigate the effect of nerve growth factor(NGF)on human vascular endothelial cell. Methods The effect of NGF on human umbilica vein vascular endothelial cell were used in the third passage.Endothelial cell proliferative rate was assessed using the enzyme linked immunoassay.The proliferating cell nuclear antigen expression and proliferating cell nuclear antigen lable index were assessed using the immunnohischemistry. Results Compared with the control group,the NGF groups whose endothelial cell proliferative rate were increased significantly(P<0.001).NGF neutralizing antibody de creased the mitogenic effect of NGF significantly(P<0.05).NGF also increased PCNA expression and PCNA lable index on HEVC304 .NGF-neutralizing antibody decreaed PCNA expression on NGF-stimulated HEVC304(P<0.05). Conclusion NGF could significantly induced the mitosis of human umbilica vein vascular endothelial cell. |
